A man and a woman, both aged 26, died in the accident, the spokesman said.
Local media earlier reported that the pair fell into a 200m ravine while driving on the mountain.
The peak is the highest point on the Greek island, which is popular with British holidaymakers.
Santorini is in the south of the Aegean Sea, southeast of the Greek capital Athens.
The British Foreign and Commonwealth Office spokesman said: “Our staff are assisting the families of two British people who have died on the island of Santorini, and are in contact with the Greek authorities.” –PA
